Minimize background bands in your Western blot by adjusting primary and secondary antibody concentrations. Sometimes, boosting blocking agent (BSA or non-fat dry milk) levels or switching to alternatives like Starting Block or SuperBlock can decrease background signal. After primary antibody incubation, wash at least twice with TBST, including 0.5 M NaCl in one or more steps. Avoid Nonidet P40 or Triton X-100 in buffers as they hinder protein detection.
